Expression CMS & ChipMate nanoESI source - two new techniques to enhance mass spectrometry

Two systems will be discussed, with application examples:
The Expression CMS is a compact, affordable mass spectrometer designed especially for chemists. Small enough to fit into a standard-sized fume hood, it allows rapid reaction monitoring by the reactor and has interface kits for flow chemistry and flash chromatography systems. It can also be interfaced to any HPLC or SFC system on the market, making it an inexpensive way to upgrade existing HPLC systems inexpensively LC/MS .

The ChipMate nano ESI source uses Advion’s proven ESI chip technology to improve the robustness and repeatability of nanoLC analyses. Uniquely, it features automated spray sensing and recovery, which allows the system to engage a new sprayer mid-run, in the event of a blockage.

An Automated On-Line Card Extraction LC/MS System for the Determination of Clozpaine and its Metabolites in Rat Blood

Dried blood spot (DBS) sampling provides several advantages when compared to conventional plasma sampling including use of small blood volumes, easy sample shipping and storage, and fewer biohazard concerns. When compared to traditional hole punching and off-line extraction methods, the on-line extraction LC/MS system enables automated flow-through DBS analysis, reduces analysis time and manual preparation, and increases sample throughput. This work describes the development and validation of an on-line card extraction DBS LC/MS method for rapid quantitation of clozapine and its metabolites, norclozapine and clozapine-N-oxide, in rat blood. Excellent sensitivity, linearity, dynamic range, precision, accuracy, and reproducibility, were demonstrated. The quantitation performance of the method is comparable to that of an off-line extraction method. This approach could be readily applied to clozapine pharmacokinetics and metabolic profiling studies.

With the globalization of the food industry, the need to monitor food quality is imminent. Mass spectrometry is a powerful tool to examine both the nutritive value of food and also food safety. We need solutions that are easy, fast, reliable and high throughput. The use of fast LC systems with high resolution time-of-flight spectrometers and software solutions to provide reliable, accurate results for food testing will be discussed. Besides the traditional analytical techniques using front end LC with electrospray ionization, we will also examine the growing field of ambient ionization in food testing. We will discuss direct sampling analysis off food surfaces with little or no sample pretreatment in complex food matrices.
